The article provides an overview of the processes leading to the generation of M0, M1, M2, and tissue-resident–like macrophages, along with their phenotypic characterization. It further discusses the metabolic programs, secretion of pro- and anti-inflammatory mediators, and functional properties associated with these cell types. Finally, the review offers a critical assessment of the use of monocyte-derived macrophages (MDMs), highlighting both their advantages and limitations.
